Sibling DNA Testing (Brother & Sister Testing)
Sometimes, families need clear answers. A sibling DNA test helps determine if two individuals share one or both biological parents.
At Purpose DNA & Health, we provide private, professional DNA testing in a calm and supportive setting.
When Is A Sibling Test Needed?
Full Siblings vs. Half Siblings – What’s the Difference?
Understanding the difference between full siblings and half siblings is important when choosing the right test and setting expectations.
Full Siblings (Same Mother and Father)
Full siblings share both biological parents. In this case, DNA testing looks for stronger genetic similarities between the individuals.
- Who should be tested?
- Two siblings (required)
- Mother (strongly recommended, if available)
- Example:
Two children believe they have the same mother and father but want confirmation. Testing both siblings, along with the mother if available, helps provide stronger and clearer results.
Half Siblings (One Shared Parent)
Half siblings share only one biological parent, either the mother or the father. These tests can be more complex because there is less shared DNA to compare.
- Who should be tested?
- Two siblings (required)
- The known parent (if available, this greatly improves accuracy)
- Example (shared father):
Two children have different mothers but believe they share the same father. Testing both siblings is required. If one of the mothers is available, including her DNA can help remove her genetic contribution and improve accuracy. - Example (shared mother):
Two children share the same mother but have different fathers. Testing both siblings and the mother can help confirm the relationship more clearly.
What If the Mother Is Not Available?
Testing can still be performed if the mother is not available, but results may be less definitive.
- The lab will rely only on the siblings’ DNA
- Results may be reported as a probability instead of a clear conclusion
- Additional family members (such as another sibling or grandparent) may help strengthen results
Important to Know
Sibling DNA testing is different from paternity testing. Because it compares shared DNA between relatives, results are based on likelihood rather than a simple yes or no. Including more known family members helps provide stronger, more confident results.

How It Works
Legal or Peace of Mind Options Available
Legal DNA testing (court-admissible)
Legal testing follows strict identification and documentation procedures to ensure results can be used in court. This option is required for cases involving child support, custody, or other legal matters.
Non-legal DNA testing (personal knowledge)
Non-legal testing provides the same accurate results but is used for personal information only. It is a simple and private option for families who want answers without legal involvement.
